Description Indirect immunofluorescence assay for the semi-quantitative detection of antibodies directed against Legionella pneumophila serogroups 1 to 6 in human serum.

Kit Components 15 slides (10 wells each) covered with a mix of inactivated Legionella bacteria, serogroups 1 to 6, Negative control, Positive control, Sample diluent, Conjugate, Mounting medium, PBS buffer.
Assay Protocol: 1. Dilute PBS buffer, and solve the sample diluant in PBS buffer. 2. Dilute the enzyme conjugate and the 2 controls in sample diluent. 3. Dilute the patient samples in sample buffer, at 1:128 for a screening, or serial dilutions for titration. 4. Add 1 drop (15 µl) of diluted samples and control to the appropriate wells, and incubate at 37°C in wet room for 30 minutes. 5. Wash the slides with diluted PBS and with distilled water. 6. Place the slides in a wet room, add 15 µl of conjugate to each well, and incubate at 37°C in wet room for 30 minutes. 7. Wash the slides with diluted PBS and with distilled water. 8. Add mounting medium and coverslip to the slide. 9. Examine the slide using a fluorescence microscope.
Storage The kit must be stored at 2-8°C.
