Description Direct fluorescent technique designed for qualitative detection of varicella-zoster virus (VZV), in vesicle smears and biopsy specimens.
Features - Use of monoclonal antibody conjugate. - Similar procedure to Merifluor FA kits.
SpecimensSmear, tissue culture, biopsy specimens.
Kit Components - Phosphate-buffered Fluorescein-labeled conjugate containing Evans Blue counterstain. - Buffered mounting medium containing photobleach inhibitor.
Specimen preparationPrepare specimens as follows: - Smear specimen: Spread collected cellular material on slide and air dry. - Tissue culture specimen: Cells demonstrating VZV cytopathic effect are dropped onto slide. - Biopsy Specimen: Frozen biopsy specimens are sectioned and air dried. Fix obtained specimen with cold acetone for 10min. Air dry. Proceed within 2H or store slide desiccated at 2-8°C.
Assay Protocol: 1. Add 1-3 drops of conjugate to specimen well. 2. Incubate at room temperature in a moist chamber for 30min. 3. Rinse carefully slides with a PBS solution. 4. Add mounting medium and apply a cover slip on the slides. 5. Read slides using a fluorescence microscope.
Storage The kit must be stored at 2-8°C.
