Description

Premier Campy is an in vitro qualitative ELISA test for the detection of specific Campylobacter antigens in stool samples from patients with signs and symptoms of gastroenteritis.  Premier Campy detects C. jejuni and C. coli in human stool that may be either unpreserved or preserved in Cary Blair-based transport media.

Specimen handling

Human stool sample that may be either unpreserved or preserved in Cary Blair-based transport media, such as Meridian ParaPak C&S (900612 – 120 vials).
Samples should be stored at 2–8 C prior to testing.  Samples should be tested as soon as possible, but may be held up to 96 hours at 2–8 C.  Samples that will not be tested within 96 hours should be frozen immediately upon receipt and stored at ≤ –20 C until tested.

 

Kit Components

Antibody coated microwells, Positive control, Negative control, Sample diluent, Wash buffer, Enzyme conjugate, Substrate, Stop solution, Transfer pipets, Strip holder, Strip sealer.

 

Assay Protocol:

1.    Emulsify 3-4 mm diameter portion of stool into 200 µl of sample diluent in a test tube, or add 50 µl of liquid stool to 200 µl of sample diluent.

2.    Transfer 100 µl of diluted stools and controls in the appropriate wells.

3.  Incubate one hour at 21-27°C (30 minutes with shaking incubation)

4.    Wash 5 times with the wash buffer.

5.    Add 2 drops of enzyme conjugate to each well, seal and incubate at 21-27°C for 30 minutes.

6.    Wash 5 times with the wash buffer.

7.    Add 2 drops of substrate to all wells, seal and incubate at room temperature for 10 minutes.

8.    Add 2 drops of stop solution and read at 450 nm, 450/630 nm or visually.

 

Storage

The kit must be stored at 2-8°C.
