Description Microplate Enzyme-Immunoassay for the qualitative detection of Rotavirus Antigen in human stool.

Specimen handlingThe stool specimen, collected in an empty container, can be stored up to 3 days at 2-8°C before testing. If testing is not performed within this time frame, specimens should be frozen upon receipt (-20°C to -80°C).
Kit Components Antibody coated microwells, Positive control, Sample diluent, Enzyme conjugate, buffered Urea Peroxide solution, Chromogen solution (TMB), Acidic Stop solution, Transfer pipets, Microwell holder.
Assay Protocol 1. Collect faecal sample by use of transfer pipette (first mark) and emulsify it into 1mL of sample diluent in a test tube. 2. Add 2 drops (100µL) of diluted stool, positive control and negative control (sample diluent) in the appropriate wells. 3. Add 2 drops (100µL) of enzyme conjugate to each well, seal and mix. 4. Incubate at room temperature for one hour. 5. Wash 5 times with deionised water. 6. Add 2 drops (100µL) of substrate to each well. 7. Add 2 drops (100µL) of chromogen solution to each well. 8. Incubate at room temperature for 10min.. 9. Add 2 drops (100µL) of stop solution. 10. Read at 450 nm or visually.
Storage The kit must be stored at 2-8°C.
