Description

Microplate Enzyme-Immunoassay for the qualitative detection of enteric Adenovirus serotypes 40 and 41 in human stools specimens.

Specimen handling

The stool specimen, collected in an empty container, can be stored up to 3 days at 2-8°C before testing.

If testing is not performed within this time frame, specimens should be frozen upon receipt (-20°C to -80°C).

 

Kit Components

Group specific anti-Adenovirus antibody coated microwells, Positive control, Sample diluent / Negative control, 40-41 serotypes specific Enzyme conjugate, Part A substrate, Part B substrate, Stop solution, Transfer pipets, Strip holder.

 

Assay Protocol:

1.    Wiith the transfer pipet, add a portion of stool into 1 ml of sample diluent in a test tube.

2.    Transfer 2 drops (100 µl) of diluted stools and undiluted controls in the appropriate wells.

3.    Add 2 drops (100 µl) of enzyme conjugate to each well, mix and incubate at room temperature for 60 minutes.

4.    Wash 5 times with deionized water.

5.    Add 2 drops (100 µl) of substrate A to each microwell.

6.    Add 2 drops (100 µl) of substrate B to each microwell, and incubate at room temperature for 10 minutes.

7.    Add 2 drops (100 µl) of stop solution, and read at 450 nm or visually.

 

Storage

The kit must be stored at 2-8°C.
