Specimen handling

Stool specimens can be either tested directly, or after culture in MacConkey broth or on a SMAC plate.

The stool specimen, collected in an empty container, can be stored up to 3 days at 2-8°C before testing. If testing is not performed within this time frame, specimens should be frozen upon receipt (-20°C to -80°C).

Refrigerated specimens should be cultured as soon as possible after receipt.

 

Kit Components

Test cards, Positive control, Negative control, Sample diluant, Transfer pipets.

 

Assay Protocol:

1.    Direct stool :

  1. Add 350 µl of sample diluent to a test tube, and transfer 25 µl of liquid stool or a small portion of solid stool (2 mm diameter) to the tube.
  2. Vortex diluted specimen for 10 seconds, then add 150 µl of diluted stool to sample port of device.
  3. Incubate 10 minutes at room temperature, and read the result of the test.

2.    Broth enrichment :

  1. Add 350 µl of sample diluent to a test tube, and transfer 350 µl of broth culture to the tube.
  2. Vortex diluted specimen for 10 seconds, then add 150 µl of diluted specimen to sample port of device.
  3. Incubate 10 minutes at room temperature, and read the result of the test.

3.    Plate culture :

  1. Add 700 µl of sample diluent to a test tube, and use a swab to make a heavy suspension of suspected growth from SMAC plate in sample diluant.
  2. Mix diluted specimen gently, then add 150 µl of diluted specimen to sample port of device.
  3. Incubate 10 minutes at room temperature, and read the result of the test.

 

Interpretation of results

Positive result : 1 band in the Control zone + 1 band in the Test zone.

Negative results : 1 band in the Control zone only.

 

Storage

The kit must be stored at 2-8°C.
