Description
One-step
single test for the qualitative detection of Shiga Toxins 1 and 2
produced by E. coli in cultures derived from clinical stool specimens.

Specimen storage
Stool specimens can be tested after overnight enrichment in McConkey broth (cat. n. 7516MC2).
Stools collected in an empty container can be stored up to 2 hours at
2-8°C before culturing. Samples in Modified Cary-Blair transport media
can be stored up to 3 days at 2-8°C before culturing.
If culturing is not performed within this time frame, specimens should be frozen at -70°C immediately upon receipt.
After enrichment, the broth may be stored for up to 7 days at 2-8°C before testing with ImmunoCard STAT! EHEC.
If testing is not performed within this time frame, the broth should be frozen at -20°C for up to 3 weeks.
Specimen enrichment in McConkey Broth
- Add 50 µl of unpreserved specimen or 150 µl of specimen preserved in
Modified Cary-Blair medium to a culture tube containing 5 ml of
McConkey broth.
- Incubate at 35-39°C for 16-24 hours.

ImmunoCard STAT! EHEC Protocol - After Broth enrichment
1. Add 150 µl of Sample Diluent to a test tube, and transfer 150 µl of broth culture to the tube.
2. Vortex diluted specimen for 10 seconds, then add 150 µl of diluted specimen to sample port of device.
3. Incubate for 20 minutes at room temperature, and read the result of the test.
Interpretation of results
Positive result: 1 band in the Control zone + 1 band in the Toxin 1, Toxin 2 or both line positions.
Negative results : 1 band in
the Control zone only.
Storage
The kit must be stored at
2-8°C.
