Description

Qualitative or semi-quantitative differential slide agglutination test for detection of infectious mononucleosis heterophile antibodies in human serum or plasma. The test is based on fresh horse erythrocytes.

Kit Components

Guinea Pig Antigen (Reagent I), Beef Erythrocyte Stroma Antigen (Reagent II), Horse Erythrocytes (Indicator cells), Positive control, Negative control, Glass Slide, Microcapillary Pipettes, Rubber Bulbs, Plastic Pipettes, Wooden Applicators.

 

Assay Protocol:

1.       Using one of the disposable microcapillary pipette, deliver 10 lambda of indicator cells to one corner of both slide squares.

2.       Place 1 drop of Reagent I in the center of square I.

3.       Place 1 drop of Reagent II in the center of square II.

4.       Place 1 drop of sample or control to the center of each square. Saline dilutions of sample can be made for title determination.

5.       Using a clean wooden applicator, mix reagent I or reagent II with sample or control, then blend in the Indicator Cells over the entire surface of each square.

6.       Observe for agglutination for no longer than 1min after final mixing.

 

Interpretation of results

Positive result : Square I agglutination pattern is stronger

Negative result : Square II agglutination pattern is stronger

 

Storage

The kit must be stored at 2-8°C.
