Description

Unique, patented system for the concentration of eggs, larvae and protozoa from preserved fecal specimens.

SpinCon is a centrifuge tube divided in two parts with two filtering screens in between. After pre-filtering the diluted stool specimens, the tube is centrifuged and the specimen is recovered in the lower part of the device. Because of the strong filtration force, there is no need for using chemical solvents, like Ethyl Acetate.

Specimens preserved in 10% Formalin, Sodium Acetate Formalin (SAF) and Ecofix may be used with the system.SpinCon

Specimen Collection

The system can be used with products from the Para-Pak line of parasitology products (10% Formalin , SAF and Ecofix) or any other collection device. The specimen must be well mixed in a preservative and must be allowed to stand for at least 30 minutes at RT prior to processing to allow adequate fixation.

If a permanently stained smear is desired, it is recommended that a portion of preserved stool be handled separately prior to concentration with SpinCon.

 

Kit Components

- SpinCon Filtration Devices: 200 or 500

- SpinCon Tubes with Caps: 200 or 500

- Para-Pak Surfactant: 2x30 ml or 1x225 ml

 

Assay Protocol:

1.              Add 2 drops of surfactant per 5 ml preserved stool specimen (8-10 drops per ParaPak vial).Shake for 60’’

2.              Filter 3 ml surfactant treated, preserved stool.

3.              Add 2 ml physiological saline to the upper half of the SpinCon for a total of 5 ml diluted, filtered stool.Cap and mix.

4.              Centrifuge  for 10 minutes at 500X g (1800-2200 rpm).Discard top half of SpinCon.

5.              Resuspend final pellet in appropriate volume of 10 % Formalin or SAF.

6.              Perform a wet mount on the resuspended sediment in saline or Lugol’s iodine and examine microscopically for the presence of parasites

Store all reagents and components at RT (15-30°C)
