Description Anti-complement Immunofluorescence assay designed for qualitative and/or semi-quantitative detection of antibodies to the nuclear antigen of Epstein-Barr Virus in human serum.

SpecimensHeat-inactivated fresh, refrigerated or frozen human serum.
Kit Components - 10-well EBV-NA substrate slides. - Lyophilised guinea pig complement.

Complement diluent. - Lyophilised Fluorescein-labeled anti-guinea conjugate: - Heat-inactivated, lyophilised human PC and NC sera. - Phosphate-buffered Saline (PBS powder). - Buffered mounting medium.
Assay Protocol: 1. Rehydrate complement with cold, sterile distilled water. 2. Prepare a 1:10 screening dilution of the heat-inactivated patient serum with rehydrated PBS. Serial dilutions can be done for title determination. 3. Reconstitute conjugate and controls with prepared PBS. 4. Add 1 drop (15µL) of diluted samples and controls to the appropriate wells. 5. Incubate at 37°C in a moist chamber for 30min. 6. Dilute rehydrated complement with the appropriate volume of cold complement diluent. 7. At the end of the incubation time, rinse slides with prepared PBS solution. 8. Add 1 drop (15µL) of cold working complement. 9. Incubate at 37°C in a moist chamber for 30min. 10. Rinse with prepared PBS solution. 11. Add 1drop (15µL) conjugate to each well. 12. Incubate at 37°C in a moist chamber for 30min. 13. Rinse with prepared PBS solution. 14. Add mounting medium and apply a cover slip on the slide. 15. Read slide using a fluorescence microscope.
StorageLyophilised Guinea pig complement must be stored at –20°C and remaining kit components at 2-8°C.
