Description Indirect immunofluorescence assay designed for qualitative and/or semi-quantitative detection of IgM antibodies to Herpes Simplex Virus in human serum.
Features - Highly specific detection of antibodies to (1+2)-HSV. - Easier and quicker procedure compared to viral isolation. - Similar procedure to all Merifluor IFA kits.
SpecimensFresh, refrigerated or frozen human serum.
Kit Components - 10-well HSV Antigen substrate slides. - Lyophilised Fluorescein-labeled anti-human IgM conjugate. - Lyophilised human PC and NC sera. - Phosphate-buffered Saline (PBS powder). - Buffered mounting medium.
Assay Protocol: 1. Prepare a 1:10 screening dilution of the patient serum with rehydrated PBS or ready-to-use IgG inactivation reagent (Gullsorb Cat.#XX715). 2. Reconstitute conjugate and controls with prepared PBS. 3. Add 1 drop (15µL) of diluted samples and controls to the appropriate wells. 4. Incubate at 37°C in a moist chamber for 60min. 5. Rinse slides with prepared PBS solution. 6. Add 1 drop (15µL) of conjugate to each well. 7. Incubate at 37°C in a moist chamber for 30min. 8. Rinse slides with prepared PBS solution. 9. Add mounting medium and apply a cover slip on the slide. 10. Read slide using a fluorescence microscope.
Storage The kit must be stored at 2-8°C.
